Mechanism of Action

Cagrilintide activates amylin receptor complexes (AMY1, AMY2, AMY3) -- heterodimers of the calcitonin receptor (CTR) and receptor activity-modifying proteins (RAMP1, 2, 3) -- expressed in the area postrema and nucleus tractus solitarius of the brainstem. Receptor activation suppresses postprandial glucagon release, delays gastric emptying, and promotes satiety through hypothalamic integration. The combination of amylin receptor agonism with GLP-1R agonism provides additive anorectic signalling through partially non-overlapping CNS circuits.
