Overview
CJC-1295 is a synthetic GHRH analogue modified with Drug Affinity Complex (DAC) technology that enables covalent binding to circulating albumin, significantly extending its plasma half-life. Ipamorelin is a selective, third-generation growth hormone secretagogue receptor (GHSR) agonist that stimulates GH release with minimal effect on cortisol or prolactin secretion.
Scientific Overview
Research has investigated this combination as a model for sustained, physiologically-patterned GH stimulation. CJC-1295 provides a prolonged GHRH signal, while Ipamorelin activates the ghrelin receptor pathway independently. Preclinical and early clinical research has examined effects on GH pulse amplitude, IGF-1 elevation, lean mass accretion, fat oxidation, and bone mineral density markers. The combination is frequently used in research exploring the synergy between two distinct GH-stimulatory pathways.
Mechanism of Action
CJC-1295 binds GHRH-R on pituitary somatotrophs, stimulating GH synthesis and release. Its DAC modification allows reactive lysine residues to form covalent bonds with albumin, extending half-life to several days. Ipamorelin acts at GHSR-1a (the ghrelin receptor) to potentiate GH release through a separate, additive pathway. Together, these two mechanisms provide complementary stimulation of the GH axis without the cortisol or prolactin elevation seen with less selective secretagogues.
