Overview
DSIP (Delta Sleep-Inducing Peptide) is a nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) first isolated from rabbit cerebral venous blood in 1977 during studies of sleep-induction activity. It is found endogenously in the hypothalamus, pituitary, and limbic system, as well as in peripheral tissues. DSIP is supplied in lyophilised form at 10mg.
Scientific Overview
Research on DSIP has explored its role in sleep architecture, stress response, and neuroendocrine regulation. Early studies demonstrated sleep-inducing properties following intracerebroventricular administration in animals. Subsequent research has examined its ability to modulate ACTH and LH release, attenuate stress-induced hormonal disturbances, and influence pain sensitivity. Its distribution across multiple tissue types and the absence of a confirmed receptor have made mechanistic characterisation an ongoing research challenge.
Mechanism of Action
The precise mechanism of DSIP remains under investigation. It has been shown to interact with NMDA and GABA-B receptor systems, which may contribute to its sleep-modulatory effects. DSIP reduces the stress-induced release of ACTH from the pituitary, potentially through modulation of CRH signalling in the hypothalamus. It has also been reported to stimulate GH release via GHRH-independent pathways. Its amphiphilic structure facilitates passage across the blood-brain barrier, and it is rapidly distributed throughout the CNS following systemic administration in research models.
