Overview
Epitalon (Epithalon; Ala-Glu-Asp-Gly) is a synthetic tetrapeptide developed by the St. Petersburg Institute of Bioregulation and Gerontology based on epithalamin, an extract isolated from the bovine pineal gland. It has been investigated primarily in the context of ageing and circadian biology, with research spanning several decades of preclinical and early clinical study.
Scientific Overview
Research on Epitalon has examined its effects on telomerase activity, melatonin synthesis, and neuroendocrine regulation. Preclinical studies have reported life-extension effects in several rodent models, associated with modulation of the hypothalamic-pituitary axis and antioxidant gene expression. Its interaction with telomere biology -- specifically, its proposed stimulation of telomerase in somatic cells -- has attracted considerable interest in longevity research.
Mechanism of Action
Epitalon has been proposed to stimulate telomerase activity in human somatic cells, an enzyme otherwise silenced in differentiated tissue, potentially contributing to telomere maintenance. It is also reported to increase pineal gland secretion of melatonin, restoring circadian rhythmicity in aged models. The peptide has been shown in preclinical research to modulate expression of p53, Rb, and antioxidant response genes, and to normalise the cortisol/melatonin ratio in aged organisms.
