Overview
Kisspeptin refers to a family of neuropeptides encoded by the KISS1 gene, which signal through the G-protein-coupled receptor GPR54 (also designated KISS1R). The biologically active forms include full-length Kisspeptin-54 and shorter processed variants (Kisspeptin-14, -13, -10), all of which share the C-terminal decapeptide required for receptor activation. Kisspeptin is supplied at 10mg in lyophilised form.
Scientific Overview
Kisspeptin is recognised as the primary regulator of gonadotropin-releasing hormone (GnRH) pulsatility and is therefore central to reproductive neuroendocrinology research. Studies have investigated its role in puberty onset, the LH surge preceding ovulation, and feedback mechanisms within the hypothalamic-pituitary-gonadal (HPG) axis. Research has also examined its role in hypothalamic control of appetite, metabolic sensing, and the integration of energy status with reproductive signalling.
Mechanism of Action
Kisspeptin activates KISS1R on GnRH neurons in the arcuate nucleus and anteroventral periventricular nucleus (AVPV), triggering membrane depolarisation via Gq/11 coupling. This leads to IP3-mediated calcium release, rapid GnRH secretion, and downstream LH and FSH release from the anterior pituitary. Kisspeptin neurons act as an integration node for sex steroid feedback: oestrogen upregulates KISS1 expression in AVPV neurons (positive feedback) while suppressing it in arcuate neurons (negative feedback), generating the cyclical hormonal dynamics essential to reproductive physiology research.
