Overview
KPV (Lys-Pro-Val) is a synthetic tripeptide derived from the C-terminal sequence of alpha-melanocyte-stimulating hormone (alpha-MSH). It represents the minimal active fragment capable of retaining the anti-inflammatory properties associated with the parent peptide, without the melanogenic activity mediated by the N-terminal sequence of alpha-MSH.
Scientific Overview
Research has investigated KPV primarily in the context of intestinal inflammation, skin biology, and wound healing. In vitro and rodent studies have demonstrated potent anti-inflammatory effects in models of inflammatory bowel disease, including colitis. KPV has been shown to suppress pro-inflammatory cytokine release from immune and epithelial cells. Its small molecular size has prompted investigation into mucosal and dermal delivery strategies within research settings.
Mechanism of Action
KPV is a partial agonist at melanocortin receptor 1 (MC1R) and potentially at MC3R and MC5R. Binding to MC1R activates intracellular cAMP signalling, which suppresses NF-kB nuclear translocation -- the central transcription factor of the acute inflammatory cascade. This reduces downstream expression of TNF-alpha, IL-1beta, and IL-6. KPV has also been shown to directly enter epithelial cells and modulate inflammatory signalling intracellularly, independent of surface receptor engagement.
