Overview
Melanotan II (MT-II) is a synthetic cyclic lactam analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), the endogenous tridecapeptide derived from POMC cleavage. It was developed at the University of Arizona as part of a research programme investigating UV-independent melanogenesis as a potential photoprotective strategy. MT-II is a non-selective agonist at melanocortin receptors MC1R through MC5R.
Scientific Overview
Research on Melanotan II has examined its effects across melanocyte biology, sexual function, appetite regulation, and CNS behaviour. Studies have demonstrated potent stimulation of melanogenesis via MC1R, leading to increased eumelanin synthesis. CNS effects mediated by MC3R and MC4R agonism have been investigated in models of sexual arousal, food intake, and energy expenditure. Research has also explored the compound's UV-protective potential through enhanced constitutive pigmentation in cell and animal models.
Mechanism of Action
Melanotan II binds MC1R on melanocytes, activating Gs-coupled adenylyl cyclase and elevating cAMP, which drives MITF (microphthalmia-associated transcription factor) expression and upregulates enzymes involved in melanin synthesis, including tyrosinase, TRP-1, and TRP-2. Concurrently, MC3R and MC4R agonism in hypothalamic nuclei modulates dopamine release in reward pathways and activates pro-opiomelanocortin neurons that suppress feeding behaviour. The non-selective receptor profile of MT-II produces broader systemic effects than selective MC1R agonists.
