Overview
MOTS-C (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino-acid mitochondrial-derived peptide encoded within the mitochondrial genome, first characterised in 2015. It represents a class of mitochondria-encoded signalling peptides with systemic metabolic effects. MOTS-C is supplied in lyophilised form at 10mg.
Scientific Overview
Research has identified MOTS-C as a regulator of metabolic homeostasis with activity in skeletal muscle, adipose tissue, and the liver. Preclinical studies have demonstrated improvements in insulin sensitivity, glucose uptake, and mitochondrial function in high-fat diet models. Circulating MOTS-C levels have been observed to decline with age and in metabolic disease states, stimulating investigation into its role in longevity and age-related metabolic dysfunction.
Mechanism of Action
MOTS-C activates AMP-activated protein kinase (AMPK) through the folate-AICAR metabolic pathway, increasing glucose uptake and fatty acid oxidation in skeletal muscle. It translocates to the nucleus under metabolic stress, where it regulates mitochondrial biogenesis gene expression. MOTS-C also modulates the methionine cycle, linking mitochondrial one-carbon metabolism to systemic metabolic regulation. Its activation of AMPK is independent of the canonical LKB1 pathway in some tissue contexts.
