Overview
MOTS-C (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino-acid mitochondrial-derived peptide encoded within the mitochondrial genome, first characterised in 2015. Scientific Overview
Research has identified MOTS-C as a regulator of metabolic homeostasis with activity in skeletal muscle, adipose tissue, and the liver. Preclinical studies have demonstrated improvements in insulin sensitivity, glucose uptake, and mitochondrial function in high-fat diet models. Circulating MOTS-C levels have been observed to decline with age and in metabolic disease, driving research into its potential longevity-related roles.
Mechanism of Action
MOTS-C activates AMP-activated protein kinase (AMPK) through the folate-AICAR metabolic pathway, increasing glucose uptake and fatty acid oxidation in skeletal muscle. It translocates to the nucleus under metabolic stress to regulate mitochondrial biogenesis gene expression. MOTS-C also modulates the methionine cycle, linking mitochondrial one-carbon metabolism to systemic regulation. Its AMPK activation is independent of the canonical LKB1 pathway in some tissue contexts.
