Overview
Nicotinamide adenine dinucleotide (NAD+) is an essential pyridine nucleotide coenzyme present in all living cells. It participates in redox reactions as an electron carrier, and serves as a co-substrate for a range of regulatory enzymes including sirtuins and poly(ADP-ribose) polymerases (PARPs). NAD+ concentrations decline significantly with age in multiple tissues. This product is supplied in lyophilised form at 500mg.
Scientific Overview
NAD+ research has expanded substantially over the past decade, with preclinical studies demonstrating its role in longevity pathways, DNA damage repair, mitochondrial biogenesis, and circadian rhythm regulation. Studies have examined NAD+ repletion strategies in the context of age-associated metabolic decline, neurodegeneration models, and cardiovascular disease. Sirtuin activation downstream of NAD+ elevation has been a primary mechanistic focus across longevity and metabolic research programmes.
Mechanism of Action
NAD+ is a co-substrate for seven sirtuin deacylases (SIRT1-7), which regulate gene expression, mitochondrial function, and stress response. It is also consumed by PARPs in the DNA strand-break repair response, and by CD38 -- a hydrolase whose activity increases with age and contributes to NAD+ decline. As an electron carrier in the mitochondrial respiratory chain, the NAD+/NADH ratio is central to oxidative phosphorylation efficiency. Elevated NAD+ activates SIRT1 and SIRT3, which deacetylate PGC-1alpha and promote mitochondrial biogenesis.
