Nicotinamide adenine dinucleotide (NAD+) is an essential pyridine nucleotide coenzyme present in all living cells. Mechanism of Action

NAD+ is a co-substrate for seven sirtuin deacylases (SIRT1-7), which regulate gene expression, mitochondrial function, and stress response. It is also consumed by PARPs in the DNA strand-break repair response, and by CD38 -- a hydrolase whose activity increases with age and contributes to NAD+ decline. As an electron carrier in the mitochondrial respiratory chain, the NAD+/NADH ratio is central to oxidative phosphorylation efficiency. Elevated NAD+ activates SIRT1 and SIRT3, which deacetylate PGC-1alpha and promote mitochondrial biogenesis.
