Overview
Pinealon (Glu-Asp-Arg; EDP) is a synthetic tripeptide bioregulator derived from the peptide fraction of the bovine pineal gland. It was developed by the St. Petersburg Institute of Bioregulation and Gerontology as part of a broader programme investigating short regulatory peptides derived from organ-specific tissue fractions. Pinealon has been investigated primarily in CNS and neuroprotective research contexts.
Scientific Overview
Research has examined Pinealon in models of age-related neurodegeneration, ischaemic brain injury, and circadian rhythm disruption. Preclinical studies report that the peptide crosses the blood-brain barrier and localises in neuronal nuclei, where it modulates gene expression. It has been associated with reduced oxidative stress markers in cortical and hippocampal tissue, increased antioxidant enzyme activity, and preservation of cognitive function in aged rodent models. Research has also explored its capacity to normalise melatonin secretion patterns.
Mechanism of Action
Pinealon is proposed to function as a chromatin-active peptide, penetrating cell nuclei and interacting with DNA-regulatory sequences to modulate transcription of antioxidant, neuroprotective, and longevity-associated genes. In pineal and cortical neurons, it has been shown to regulate expression of antioxidant enzymes (catalase, SOD) and reduce lipid peroxidation. Its ability to cross the blood-brain barrier -- attributed to its small size and lipophilicity -- allows direct access to CNS tissue without the need for carrier systems in preclinical research models.
