Overview
Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a synthetic heptapeptide analogue of the endogenous immunomodulatory tetrapeptide Tuftsin (Thr-Lys-Pro-Arg), with an appended Pro-Gly-Pro sequence that confers enzymatic stability. It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences and has been investigated for anxiolytic, nootropic, and immunomodulatory properties.
Scientific Overview
Research has examined Selank's effects on anxiety behaviour, cognitive function, and neuroimmune regulation. Rodent studies have demonstrated anxiolytic effects comparable to benzodiazepines without associated sedation or dependence in preclinical models. Investigational work has also explored its role in BDNF upregulation, enkephalin metabolism, and modulation of the IL-6 cytokine system. Its capacity to enhance learning and memory consolidation in stressed models has been a recurring area of preclinical interest.
Mechanism of Action
Selank inhibits enzymes responsible for degrading enkephalins -- endogenous opioid peptides that modulate GABAergic transmission -- thereby increasing their availability in synaptic clefts. It modulates the GABAergic system to produce anxiolytic effects without benzodiazepine receptor binding. Selank also upregulates BDNF and its TrkB receptor in limbic structures, potentially supporting synaptic plasticity and memory processes. Immunologically, it modulates IFN-gamma and IL-6 expression, consistent with the immunomodulatory activity of its parent peptide, Tuftsin.
