Overview
Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the ACTH(4-7) sequence, to which a Pro-Gly-Pro C-terminal extension has been added to confer resistance to enzymatic degradation. Unlike native ACTH, Semax does not possess adrenocorticotropic activity. It was developed in Russia and has been investigated primarily for neuroprotective and cognitive research applications.
Scientific Overview
Research has examined Semax in the context of neurotrophic factor modulation, neuroprotection, and cognitive function. Preclinical studies have demonstrated upregulation of brain-derived neurotrophic factor (BDNF) and its receptor TrkB following Semax administration. Research in ischaemia models has shown reduced infarct volume and improved neurological outcomes. Studies have also investigated its effects on dopaminergic and serotonergic transmission, attention, and learning paradigms in rodent models.
Mechanism of Action
Semax increases BDNF expression in the cortex, hippocampus, and basal forebrain via activation of the MAP kinase (ERK1/2) signalling cascade and CREB phosphorylation. Elevated BDNF supports synaptic plasticity, neuronal survival, and axonal branching. Semax also modulates the serotonergic and dopaminergic systems by influencing monoamine transporter expression and catecholamine turnover in prefrontal regions. In ischaemia models, it reduces oxidative damage to neurons by enhancing endogenous antioxidant enzyme expression and suppressing NF-kB-mediated inflammatory gene induction.
