Overview
SS-31 (also known as Elamipretide or MTP-131) is a synthetic, cell-permeable aromatic-cationic tetrapeptide with the sequence D-Arg-2'6'-dimethylTyr-Lys-Phe-NH2. It was designed to selectively localise to the inner mitochondrial membrane, where it interacts with cardiolipin, a phospholipid critical to mitochondrial cristae architecture and respiratory chain function.
Scientific Overview
SS-31 has been investigated as a mitochondria-targeted research tool for conditions involving mitochondrial dysfunction, oxidative stress, and cristae remodelling. Research has examined its effects in cardiac ischaemia-reperfusion models, skeletal muscle ageing, and renal tubular injury. Clinical investigation has explored its potential in heart failure with preserved ejection fraction (HFpEF) and Barth syndrome, a rare cardiolipin-related mitochondrial disease.
Mechanism of Action
SS-31 rapidly accumulates in the inner mitochondrial membrane via electrostatic attraction to the negatively charged phospholipid cardiolipin. This interaction stabilises cristae structure, optimises the spatial organisation of ATP synthase dimers, and improves electron transport chain coupling efficiency. By reducing cardiolipin peroxidation, SS-31 decreases electron leak and superoxide generation from Complex I and III, suppressing mitochondrial reactive oxygen species (ROS) without directly scavenging them.
