Overview
TB-500 is a synthetic analogue of the C-terminal region of Thymosin Beta-4 (TB4), a naturally occurring 43-amino-acid actin-sequestering protein expressed in virtually all nucleated mammalian cells. The synthetic fragment studied under the designation TB-500 retains the core actin-binding domain. It is supplied as a lyophilised powder.
Scientific Overview
Preclinical and in vitro research has investigated TB-500 in the context of wound healing, muscle regeneration, cardiac repair, and corneal epithelial recovery. Studies have examined its effects on cell migration, angiogenesis, and the inflammatory response following acute tissue injury. Its role in upregulating matrix metalloproteinases and promoting endothelial tube formation has been a particular area of investigation.
Mechanism of Action
TB-500 binds monomeric G-actin and promotes actin polymerisation dynamics through interaction with the Wiskott-Aldrich syndrome protein (WASP) family. It has been shown to upregulate VEGF expression, stimulating endothelial cell proliferation and new vessel formation. Additionally, the peptide downregulates inflammatory cytokines in wound models and promotes the directional migration of keratinocytes, fibroblasts, and endothelial progenitor cells via Rho/Rac1 pathway modulation.
