Overview
Tesamorelin is a synthetic analogue of growth hormone-releasing hormone (GHRH) in which the native 44-amino-acid sequence is stabilised by the addition of a trans-3-hexenoic acid group at the N-terminus. This modification substantially extends its plasma half-life relative to endogenous GHRH while preserving receptor binding affinity.

Mechanism of Action
Tesamorelin binds to GHRH receptors on somatotroph cells in the anterior pituitary, stimulating pulsatile release of endogenous growth hormone. This in turn elevates circulating IGF-1 and IGF-binding protein-3. The resulting anabolic and lipolytic signalling cascade reduces visceral adipose tissue accumulation. Unlike direct GH administration, endogenous pulsatile release is preserved, which is considered to offer a more physiological secretion pattern in research models.
